Effect of a new inhibitor of cholesterol biosynthesis (AY 9944) on serum and tissue sterols in the rat.
نویسنده
چکیده
AY 9944 is a novel inhibitor of cholesterol biosynthesis which appears to act on the Delta 7-reductase enzyme, interfering with the conversion of 7-dehydrocholesterol to cholesterol. In all tissues studied, with the exception of the lens, there was a reduction in cholesterol and a marked accumulation of 7-dehydrocholesterol. Isotope studies showed a reduced uptake of acetate into cholesterol, and an increased uptake into 7-dehydrocholesterol, in various tissues in vivo and by skin samples in vitro. There was no apparent accumulation of lanosterol or of the methostenols in the rat skin.

Normal and inhibited cholesterol synthesis in the cultured rat embryo.
The Smith-Lemli-Opitz syndrome-affected fetus presents a deficiency in delta7-dehydrocholesterol reductase, the last enzymatic step in the cholesterol biosynthesis pathway. Development of the abnormal human fetus takes place in a normal environment as the heterozygous mother's cholesterolemia remains normal. An animal model for this disease has been obtained from the offspring of pregnant rats ...
متن کاملBrain sterols in the AY-9944 rat model of atypical absence seizures.
PURPOSE The AY-9944 (AY)-treated rat is a reproducible and clinically relevant animal model of atypical absence seizures. AY inhibits cholesterol synthesis, but the relation between brain sterol levels and the spontaneously recurrent absence seizures has not been determined. METHODS Long-Evans hooded rats were treated every 6 days from postnatal day (P)2 to P20 with AY (7.5 mg/kg, s.c.) or sa...

1. The effect of inhibition of cholesterol synthesis by a hypocholesterolaemic drug (AY-9944) was studied in rat brain during development. 2. At 2 weeks after administration of AY-9944 to young rats 7-dehydrocholesterol accounted for half the total sterol of myelin and other subcellular components. 3. At 4 weeks after injection of the drug 7-dehydrocholesterol had disappeared whereas the choles...
متن کاملLimb malformations of rat fetuses exposed to a distal inhibitor of cholesterol biosynthesis.
Triparanol, an inhibitor of desmosterol Delta24 reductase, produces a high rate of limb malformations in rat fetuses exposed at gestational day 10 (gd 10) to a single oral dose (150-200 mg/kg) given to the pregnant dam. AY9944, another efficient distal inhibitor of cholesterol biosynthesis that blocks dehydrocholesterol Delta7 reductase, produces a similar degree of cholesterol depletion but fe...
متن کاملAn Ultrastructural and Biochemical Study of the Effects of Three Inhibitors of Cholesterol Biosynthesis upon Murine Adrenal Gland and Testis
Triparanol and 20,25-diazacholesterol inhibit cholesterol biosynthesis and result in the accumulation of desmosterol. AY-9944, another inhibitor, produces an accumulation of 7-dehydrocholesterol. Adult male C3H mice receive one of these drugs intraperitoneally. Livers, adrenal glands, and testes from each drug group are excised, and portions of each are analyzed by a modified Liebermann-Burchar...
